This groundbreaking achievement is the result of Colossal Biosciences, a biotechnology firm located in Texas, USA. On April 8, 2025, the company introduced three cloned Direwolf pups: two males named Romulus and Remus, and a female named Khaleesi. These names, drawn from mythology and popular culture, emphasize the remarkable resurgence of this creature.

Utilizing Ancient DNA and Advanced Technology

The de-extinction process involved sophisticated gene editing and cloning methodologies. Scientists retrieved DNA from Direwolf fossils—a tooth estimated to be 13,000 years old and a skull believed to be 72,000 years old. This ancient DNA was utilized to alter the blood cells of contemporary gray wolves, their closest living relatives. The modified genetic material was then inserted into the egg cells of domestic dogs, which were subsequently gestated by surrogate mothers.

A Landmark Achievement in Science

Ben Lamm, the CEO of Colossal, described the project as a “historic scientific achievement.” This endeavor represents one of the most advanced applications of synthetic biology in the field of de-extinction.

What Made Direwolves Unique?

The Direwolf was not just an ordinary predator. Although comparable in length to today’s largest gray wolves, it was significantly more powerful, weighing between 60 and 68 kilograms. With a broader skull, shorter legs, and stronger jaws, it was exceptionally equipped to hunt large prey such as bison, horses, camels, and even mastodons.

A Distinct Lineage

Genetic studies conducted in 2021 revealed that Direwolves are genetically separate from modern canines, leading scientists to categorize them under the genus Aenocyon, which means “terrible wolf” in Ancient Greek.
